Ornidazole Pharmacodynamics
Antimicrobial and antiprotozoal drug.
Ornidazole is effective against Trichomonas vaginalis, Entamoeba histolytica, Giardia lamblia (Giardia intestinalis) as well as some anaerobic bacteria such as Bacteroides spp. and Clostridium spp., Fusobacterium spp., and anaerobic cocci: Peptostreptococcus spp.
Aerobic microorganisms are not sensitive to ornidazole.
Indications
Trichomoniasis (urogenital infections in women and men caused by Trichomonas vaginalis); amebiasis (all intestinal infections caused by Entamoeba histolytica, including dysentery, and all extraintestinal forms of amebiasis, especially liver abscesses); giardiasis.
Prophylaxis of anaerobic infections during colorectal and gynecologic surgeries.
Contraindications
Hypersensitivity to ornidazole or other ytroimidazole derivatives, components of the drug. Children under 3 years of age and body weight less than 20 kg.
I trimester of pregnancy, period of breast-feeding.
Organic diseases of the central nervous system.
Pathological changes in the blood and abnormalities of blood cells
With caution:
Liver disease, alcoholism, pregnancy II and III, children over 3 years of age and body weight greater than 20 kg, elderly patients.
Pregnancy and lactation:
Ornidazole is contraindicated in the first trimester of pregnancy and during breastfeeding. In experiments ornidazole has no teratogenic or toxic effect on the fetus. As there have been no controlled trials in pregnant women, it is possible to use ornidazole in II and III trimesters of pregnancy only in case of absolute indications, when possible benefits of its usage for a mother exceed potential risk for a fetus.
Dosage and administration regimen
- Ornidazole is taken orally after a meal with plenty of water.
- Trichomoniasis
Adults and children over 35 kg 1 tablet 2 times a day (morning and evening) for 5 days.
To eliminate the possibility of reinfection, the sexual partner in all cases should receive the same course of treatment. - The daily dose for children with a weight of 20-35 kg is 25 mg/kg of body weight and is administered in a single dose, for 5 days.
- Amoebiasis
Treatment regimens: a three-day course for patients with amebic dysentery and a 5-10-day course for all forms of amebiasis. - Duration of treatment
- Daily dosage
Adults and children with body weight over 35 kg Children with body weight 20-35 kg
a) 3 days, 3 tablets in one dose in the evening. For body weight over 60 kg, 4 tablets (2 tablets in the morning and 2 tablets in the evening)
40 mg/kg of body weight at one sitting
b) 5-10 days 2 tablets (1 tablet in the morning and in the evening) 25 mg/kg of body weight at a single dose. - Lambliasis
Adults and children with body weight of more than 35 kg: 3 tablets once in the evening. - Children with body weight of 20-35 kg: 40 mg/kg once a day. The duration of treatment is 1-2 days.
- Prophylaxis of anaerobic infection during surgeries
The dose is 0.5-1 g 1-2 hours before surgery, and after surgery – 0.5 g 2 times a day for 3-5 days.
