Paracetamol Pharmacodynamics
Paracetamol contained in suppositories has antipyretic and analgesic effects. The drug blocks cyclooxygenase in the CNS, affecting the centers of pain and thermoregulation. The loss of body heat in febrile states increases as a result of vasodilation and increased peripheral blood flow. In inflamed tissues, cellular peroxidases neutralize the effect of paracetamol on cyclooxygenase, which explains the almost complete lack of anti-inflammatory effect of the drug.
Indications
Pain syndrome of mild to moderate intensity of different genesis (including headache, migraine, toothache, neuralgia, myalgia, algodysmenorrhea, pain in trauma, burns). Fever in infectious and inflammatory diseases.
Contraindications .
Chronic active alcoholism, hypersensitivity to paracetamol.
Dosage and administration
- Orally or rectally in adults and adolescents weighing more than 60 kg, a single dose of 500 mg, up to 4 times / maximum therapy duration – 5-7 days.
- Maximal doses: single dose 1 g, daily dose 4 g.
- Oral doses for children aged 6-12 years – 250-500 mg, 1-5 years – 120-250 mg, 3 months to 1 year – 60-120 mg, up to 3 months – 10 mg/kg. Single doses in rectal administration in children aged 6-12 years – 250-500 mg, 1-5 years – 125-250 mg.
- Frequency of use is 4 times at an interval of at least 4 hours. Maximum duration of treatment – 3 days.
- Maximum dose: 4 single doses per day.
