Nitroxoline Pharmacodynamics
Antimicrobial agent of broad spectrum action. Nitroxoline selectively inhibits bacterial DNA synthesis and acts both on Gram-positive microorganisms: Staphylococcus spp. (including beta-haemolytic streptococci, Streptococcus pneumoniae, Enterococcus faecalis), Corynebacterium diphtheriae Bacillus subtilis; and Gram-negative microorganisms: N. gonorrhoeae, E. coli, Proteus spp, Klebsiella spp., Salmonella spp., Shigella spp., Enterobacter spp. Nitroxoline is also active against Mycobacterium tuberculosis, Trichomonas vaginalis, some types of fungi (Candida spp., dermatophytes, molds, some agents of deep mycoses).
Indications
Infectious and inflammatory diseases of mainly urogenital tract (including pyelonephritis, cystitis, urethritis, epididymitis; infected adenoma or carcinoma of the prostate, etc.), caused by microorganisms sensitive to nitroxolin.
Prevention of infectious complications during diagnostic and therapeutic manipulations (catheterization, cystoscopy) in the postoperative period during kidney and urinary tract surgery.
Contraindications
Renal dysfunction accompanied with oligo- or anuria; severe liver diseases; glucose-6-phosphate dehydrogenase deficiency; children under three years old (taking into account the solid indivisible dosage form), cataract, neuritis, polyneuritis, pregnancy (III trimester), lactation. Hypersensitivity to drugs of quinoline series.
Directions for use and dosages.
- The drug is taken orally.
- The daily dose for adults is 400-800 mg.
- Frequency of intake of 3-4 times a day at intervals of 6-8 hours. The course of treatment is 2-3 weeks.
- In chronic urinary tract infections the drug can be administered repeatedly for 2 weeks with a 2-week break.
- The average daily dose for children over 5 years old is 0.2-0.4 g (0.05-0.1 g four times daily), under 5 years old – 0.2 g daily.
- For prevention of infection during kidney and urinary tract surgeries are prescribed 0.1 g per administration 4 times a day for 2-3 weeks.
- Children are prescribed in a daily dose of 10-30 mg/kg in 3-4 doses.
